ANTIOXIDANT ACTIVITY IN SERUM AND LIVER HOMOGENATE OF NORMAL AND MONOSODIUM GLUTAMATE INTOXICATED RATS CO-TREATED WITH ARTEMETHER-LUMEFANTRINE

Monosodium glutamate (MSG or White Maggi) is one of the world's most extensively used food additives which are ingested as part of commercially processed foods.It is taken as a flavor enhancer.

The manufacturers of MSG or White Maggi maintain that it is safe for human consumption,On the contrary various research reports indicated MSG-Induced toxic influences in animals.

Malaria is a parasitic disease, spread by mosquitoes and it affects millions of people worldwide thereby causing significant illness and mortality.The parasite has become resistant to a number of previously effective drugs, and so combinations of drugs are used to try increase cure and to prevent further resistance.

Artemether-lumefantrine is one such drug combination.

Superoxide dismutases are enzymes that function to catalytically convert superoxide radical to oxygen and hydrogen peroxide.

Aim and objectives of the study

The aim was to determine the superoxide dismutase activity in the serum and liver homogenate of normal and monosodium glutamate intoxicated male cluster albino rats co-treated with artemether-lumefantrine.

The objectives were to determine superoxide dismutase activity following combined administration of varied doses of Artemether-Lumefantrine in normal and MSG treated rats.

Justification of study

  • MSG is widely used flavorant reported to induce oxidative stress, hence there is great need to investigate MSG toxicity and proffer solution in regards to food and drug interaction,especially anti-malaria drugs (artemether-lumefantrine).

A whole lot of references are required here but because the @research work publication is in progress, I will be giving you my journal address in my next article to enable you access all duly consulted works.
This is just a tip of the iceberg.

Experimental Design

The experimental rats were weighed after 14 days acclimatization and randomly sectioned into six (6) groups of five (5) rats each. Administration of artemether-lumefantrine and MSG was done with the aid of a gavage and daily for 7days.

  • Group 1 (control) : They were fed rat chow (vital feed pellets) and portable tap drinking water only.

  • Group 11 (Therapeutic dose): rats were administered with the therapeutic dose, of the artemether-lumefantrine drug with regard to an average 70kg man,that is,1.62g for 4tablets,which were given according to the corresponding weight of the rats(0.0026g of the drug that gives 0.16ml) and were also given vital feed and water.

  • Group 111(Overdose): Overdose was calculated as therapeutic dose multiplied by 5,that is, 0.8ml.Also given vital feed and water.

  • Group 1V: A known toxicant group (monosodium glutamate at 8mg/g body were administered to the rats only, were also given vital feed and water.

  • Group V: Therapeutic dose (0.17ml) + MSG at 8mg/g body weight, also fed with vital feed and water.

  • Group V1: Overdose (0.85)+MSG at 8mg/g body weight.

Conclusion

It can be concluded that the therapeutic or normal dose of artemether-lumefantrine elicited a significant in-vitro antioxidant property and may protect against MSG-induced oxidative stress by increasing superoxide dismutase activity.The overdose of artemether-lumefantrine + MSG, markedly increased studied oxidative stress indicator of the rats, hence such combination use in animals (human beings too) should be discouraged.

Recommendation

  • Self medication especially antimalarial drug with the active ingredients artemether-lumefantrine should be avoided, but if it is to be administered should be at its recommended therapeutic dose.

  • MSG or white Maggi is a widely used flavour enhancer reported to induce oxidative stress, it is then recommended for people that want to take antimalarial drug should go for the one with the active ingredients artemether-lumefantrine as it tend to protect against MSG-induced oxidative stress by lowering free radical action and increase antioxidant capacity.
