praying in the Holy Spirit simply means praying in tongues. It is a missing act in the Church today.
Praying always with all prayer and supplication in the Spirit, and watching thereunto with all perseverance and supplication for all saints;Ephesians 6:18
Praying in the Holy Spirit is a very effective form of prayer; it helps us pray target prayers. This is because it is the Holy Spirit praying through us. The Bible says:
For what man knoweth the things of a man, save the spirit ofman which is in him! even so the things of God knoweth no man, but the Spirit of God.I Corinthians 2:11
This form of prayer is an advantage for the New Testamen Church, because we have the Holy Spirit dwelling tn us. Now imagine the kinds of victory we should expect in prayers we engage the Holy Spirit in praying! We would surpass Elijah and Daniel in results!
Praying The Will Of God
Whenever you pray in tongues what you are doing is praying the of God. The Holy Spirit is the Spirit of God and so knows God's will on every matter So when you engage Htm in prayer, He helps you to pray the will of the Father on that issue. That is what I call, "Target Praying", as you can't miss. And when we pray according to HIS will, we are guaranteed speedy answers (l Jn. 5:14-15).
Likewise the Spirit also helpeth our infirmities: for know not what we should pray for as vee ought: but the Spirit itself maketh intercession for us with groanings which cannot be uttered.
And he that searcheth the hearts knoweth what is the mind of the Spirit, because he maketh intercession for the saints according to the will of God. Romans 5:26-27

The Holy Spirit makes intercessions on our behalf. But you must be truly in the spirit to be able to pray effectively in the spirit. You do this by engaging your spirit man in praying not just your intellect. When this is done, you are communicating at heaven's frequency, and so are bound to HOLY outstanding results. No wonder the devil wearies people and tries to stop them from praying in the spirit. Not only does he not understand what is being said and so can't hinder your prayers and answers, but he also doesn't like the fact that you get instant results in prayers. That's why you must violently break forth praying in the spirit on issues that have refused to go.
May be that issue has refused to go because you do not yet have or know the will of God that will commit Him to respond. Well, that is taken care of when you pray in tongues, because the Holy Spirit that knows the will of God goes straight to tackle the matter, using the will of God! He knows what is in the mind of God, so He prays according to the Will of God on your behalf to set you free! No wonder Apostle Paul's prayers were always being answered. He was a man that prayed in the Spirit very often; he said he prayed in the spirit more than everyone else (l COL 14:14-18).

Praying in the Holy spirit builds our faith, it makes us to concentrate on God. It make us to confuse the enemy, because he that prayeth in an unknown tongue prayeth not to man but unto God.
It is a spiritual boost in our spirit. Praying in the spirit makes us to pray accordingly to the will of God.
It is a sign of the Holy Ghost if deception is out of it.

The holy spirit helps us pray target prayers, God has filled the earth with the holy spirit to be our guide.
Prayer is, for the most part, an untapped resource, an unexplored continent where untold treasure remains to be unearthed. It is talked about more than anything else , and practiced less than anything else. And yet, for the believer it remains one of the greatest gift our Lord has given us outside of salvation.
The Holy spirit is our comforter. We as christians praying for Holy spirit is just like praying for guidance. As the Gift of the holy spirit comes with many good benefits
that divine attributes are also ascribed to the #HOLY_SPIRIT, omnipresence (Ps. 139:7; Eph. 2:17, 18; 1 Cor. 12:13); omniscience (1 Cor. 2:10, 11); omnipotence (Luke 1:35; Rom. 8:11); eternity (Heb. 9:4).
Creation is ascribed to #him (Gen. 1:2; Job 26:13; Ps. 104:30), and the working of miracles (Matt. 12:28; 1 Cor. 12:9-11).
Worship is required and ascribed to #him (Isa. 6:3; Acts 28:25; Rom. 9:1; Rev. 1:4; Matt. 28:19).

Praying in the Holy Spirit is the ability given to believers that allows them to pray in a supernatural language. It is also sometimes referred to as praying in tongues, praying in unknown tongues, or praying with your spirit language. The Holy Spirit gives us power and boldness and gives us what to say in prayers when we're short of words.
Just as @mycar said, praying in the will of God is also very vital. You can't pray amiss and expect answers.
